Ultrastructural characteristics of human oocytes vitrified before and after in vitro maturation

چکیده
The development of an effective program that combines in vitro maturation (IVM) and cryopreservation for immature oocytes would represent a novel advance for in vitro fertilization (IVF), especially as a means to preserve the fertility of women in unique situations. The aim of this study was to analyze the ultrastructural characteristics of human oocytes, obtained after controlled ovarian stimulation, to determine whether IVM is best performed before or after vitrification. To this end, we analyzed the following features in a total of 22 MII oocytes: size, zona pellucida and perivitelline space, mitochondria number, M-SER (mitochondria-smooth endoplasmic reticulum) aggregates and M-V (mitochondria-vesicle) complexes, the number of cortical granules and microvilli, and the presence of vacuolization using transmission electron microscopy (TEM). Each oocyte presented a rounded shape, with an intact oolemma, and was surrounded by a continuous zona pellucida and perivitelline space. Statistical analysis comparing oocytes vitrified before or after IVM indicated that there were no significant differences between examined characteristics.

Purpose: Our aim was determination of the sheep oocytes ultrastructural changes follow vitrification and in vitro maturation. Materials and Methods: Good quality isolated cumulus-oocyte complexes (COCs) were randomly divided into non-vitrified control, conventional straw, cryotop and solid surface vitrification groups. In the conventional and cryotop methods the vitrified COCs were plunged dire...
متن کاملVitrification of human germinal vesicle oocytes; before or after in vitro maturation?
Background The utilization of immature oocytes derived from stimulated cycles could be of great importance, specially for urgent fertility preservation cases. The aim of the current study was to determine whether in-vitro maturation is more successful before or after the vitrification of these oocytes. MaterialsAndMethods The study was performed in a private IVF center. 318 germinal vesicle (GV...
متن کاملP-145: Maturation Capacity and Viability Assessment of Human Immature Oocytes After Vitrification and In Vitro Maturation (IVM)
Background: 15% of oocytes collected in ART cycles are immature. These oocytes may be matured following IVM program. It is possible to cryopreserve the immature oocytes for further use in ART after application of IVM. The aim was to determine the maturation rate, morphology and viability of human immature oocytes after fresh IVM and vitrified-IVM program. Materials and Methods: 63 women who und...
متن کاملP-92: Specific Activation Requirements of In Vitro Matured Sheep Oocytes following Vitrification-Warming
Background: Oocyte vitrification and assisted oocyte activation have increasingly important implications in assisted reproductive technology. However, an important area of concern with matured oocyte cryobiology is that elements of oocytes intimately involved in metaphase-II arrest may be modified by cryopreservation. In a comparison between different cellular characteristics of unvitrified, vi...
متن کاملI-14: Vitrification of Human Immature Oocytes in IVM Program
The cryopreservation of immature GV oocytes by vitrification is offering attractive perspectives, since they represent an important pool of germ cells for women undergoing IVM cycles, or at risk of infertility for OHSS or PCOs, particularly those poor responders to gonadotropin stimulation, or in case of cancer. Therefore, it becomes necessary to optimize both cryopreservation technique as well...
